Study on hypoglycemic activity of purified polysaccharide from Inonotus obliquus in vitro

Insulin resistance is the common pathophysiological basis and common risk factors of many metabolic related diseases, which has become the central link of type II diabetes. Therefore, we can start from the prevention and treatment of the disorder of glucose and lipid metabolism to find excellent new drugs for diabetes from traditional Chinese medicine. In this paper, human HepG2 cells were used to study the glucose consumption effect of Inonotus obliquus polysaccharide on IR HepG cells of normal HepG2 cells. The experimental results showed that the purified Inonotus obliquus polysaccharide could significantly promote the glucose consumption of IR HepG2 cells induced by hyperinsulinemia, and significantly increase the glucose consumption of IR HepG2 cells, which was superior to metformin. The results showed that the purified polysaccharide of Inonotus obliquus could improve the insulin resistance and enhance the uptake and utilization of glucose by cells, which was consistent with the research results of Longkai and Li Changgui. High concentration (80 μ g / ml) of eiop1 can reduce the glucose consumption of IR HepG2 cells, while the same concentration of eiop2 can keep the same effect on glucose consumption. The reason may be closely related to its physical and chemical properties, monosaccharide composition, molecular weight and structural characteristics. The results also showed that the glucose consumption of HepG2 and IR HepG2 cells could be significantly promoted by two kinds of Inonotus obliquus polysaccharides within a certain concentration range, and different concentrations of Inonotus obliquus had different effects on glucose consumption. Follow-up research can focus on the effect of Inonotus obliquus polysaccharides on the activity of key enzymes in glucose metabolism. In conclusion, the α - glucosidase inhibitory activity of the purified two polysaccharides is significantly higher than that of the crude polysaccharide, and the effect of promoting glucose consumption of cells in vitro is not the same as that of the crude polysaccharide, which may be due to the complexity of polysaccharide components in the crude polysaccharide of Inonotus obliquus and the synergistic effect between them. The specific synergistic mechanism needs to be studied.
