Vinegar is a seasoning commonly used on the table since ancient times. Vinegar is produced by acetic acid fermentation of alcohol, and its metabolism is through ADH and ALDH pathway. The manufacturing process of vinegar is similar to that of alcohol metabolism of human body. Perhaps the components with ADH and ALDH activity produced in the manufacturing process of vinegar can promote alcohol decomposition and reduce alcoholic liver injury. In this study, "Acetobacter powder" was used as the research object to establish an animal experimental model to evaluate its effect on liver lipid accumulation and oxidative damage caused by acute alcohol intake, so as to provide a theoretical basis for the development of a new type of nutritional auxiliary food to alleviate alcoholic liver damage.
Tian Simin, Chen Xiaoxue and Han Beizhong from China Agricultural University evaluated the effect of Acetobacter powder on liver lipid accumulation and oxidative damage caused by acute alcohol intake. 25 male ICR mice were randomly divided into control group, ethanol group, Acetobacter powder high (45 mg / kg), medium (15 mg / kg), low (7.5 mg / kg) dose group. The contents of TG, GSH and MDA in the liver of mice in each group were measured. The activities of ADH and ALDH in Acetobacter powder were analyzed. The results showed that compared with the ethanol group, the TG concentration of the high dose group was significantly lower (P & lt; 0.05), the GSH concentration was significantly higher (P & lt; 0.01), the MDA content was not significantly changed (P & gt; 0.05), and the degree of fatty degeneration was reduced; the ADH and ALDH enzyme activities of Acetobacter powder were 0.48 U / Mg and 1.38 U / Mg, respectively. In conclusion, Acetobacter powder can reduce the liver lipid accumulation and oxidative damage caused by acute alcohol intake.
There was no significant difference in the body mass between each dose group and ethanol group (P & gt; 0.05). This shows that the body weight difference of mice used in the experiment has no significant effect on the follow-up results.
The TG level and oil red O staining of the liver were statistically analyzed. Compared with the control group, the TG concentration of the liver in the ethanol group was significantly higher (P & lt; 0.01), which could be used to determine the success of the model. The TG level of Acetobacter powder group was significantly lower than that of ethanol group (P & lt; 0.01), and the TG level of 45 mg / kg group was almost the same as that of blank control group.
The formation of lipid droplets in the liver of mice in the control group was less, and the cells were scattered; the liver of mice in the ethanol group was diffuse steatosis, most of the hepatocytes were filled with lipid droplets with different sizes and uneven distribution of vacuoles, some of them were fused and distributed in the hepatocyte cords, and the distribution and range of lipid droplets in the liver were higher than those in the control group; 45 In mg / kg group, the degree of fat denaturation was reduced, and the vacuoles of fat droplets were smaller, but they were still diffuse. According to the histological analysis, there was lipid accumulation in the liver of mice in the ethanol group. Compared with the ethanol group, the degree of lipid accumulation in the liver of mice in the 45 mg / kg group was lighter, which indicated that the Acetobacter powder could alleviate the lipid accumulation caused by alcohol.
Compared with the control group, the concentration of MDA and GSH in the liver of the ethanol group was significantly higher (P & lt; 0.05), and the concentration of GSH was significantly lower (P & lt; 0.05), which can be used to judge the success of the model. The GSH level of 15 mg / kg group and 45 mg / kg group was significantly higher than that of ethanol group (P & lt; 0.05), and there was no significant difference in MDA level between each dose group and ethanol group (P & gt; 0.05).
The regression equation of the standard curve of the activity and effect of Acetobacter powder enzyme was y = 0.1947x-0.0027, and the linearity was good (R2 = 0.9994). The ADH and ALDH activities of Acetobacter powder were 0.48 U / Mg and 1.38 U / Mg, respectively. The accumulation of liver fat caused by ethanol can be reduced by feeding Acetobacter powder at the same time of single feeding.
conclusion
By simulating acute alcohol intake, an animal model was established to evaluate the effect of Acetobacter powder on liver lipid accumulation and oxidative damage caused by acute alcohol intake. The intake of Acetobacter powder can reduce the liver lipid accumulation and oxidative damage caused by acute alcohol intake.
